Development of Amorphous Drug Nanoparticle-Forming Solid Dispersion Formulations

本研究では,薬物非晶質ナノ粒子形成型固体分散体製剤(Amorphous solid dispersion: ASD)の開発および薬物非晶質ナノ粒子形成が薬物吸収性に及ぼす影響の定量的評価を行った.難水溶性モデル薬物としてfenofibrate(FFB),ポリマー担体としてhypromellose(HPMC)を用い,噴霧乾燥によりFFB/HPMC ASDを調製した.FFB/HPMC物理的混合物(Physical mixture: PM)懸濁液中では,μmオーダーのFFB結晶が分散していた一方,FFB/HPMC ASD懸濁液では非晶性の約160 nmのナノ粒子の形成が認められた.FFB/HPMC ASD懸濁液中に溶解したFFB濃度は,FFB/HPMC PM懸濁液と比較して約8.6倍高い値を示した.ラット小腸上皮細胞へのナノ粒子の取り込みを評価した結果,ナノサイズの非晶質FFB粒子が直接小腸に取り込まれることが示唆された.FFB/HPMC ASD懸濁液からのFFB消化管吸収速度はFFB/HPMC PM懸濁液と比較して約38倍改善し,FFB溶解量の改善度(約8.6倍)を上回るFFB吸収性改善が認められたことから,非晶質FFBナノ粒子の小腸上皮細胞における直接取り込みが顕著なFFB吸収改善に寄与したと結論付けた.
